What is Doxepin?
Doxepin is an active ingredient used for the treatment of anxiety, depression and major depressive disorder. The product ingredient is Doxepin hydrochloride (hcl).
The brand names in the United States are: Silenor, Sinequan, Adapin.
Mechanism of Action (MOA)
It blocks the reuptake of neurotransmitters by the neuronal membrane.
Therapeutic Indications, Uses and Benefits of Doxepin
- Psychoneurotic disorders where anxiety and/or depression are prominent symptoms: neurosis with anxiety with or without somatic symptoms, reactive depression, mixed anxiety depression.
- Psychotic depressions including involutive melancholy and depressive phase of manic depressive psychosis.
Dosage of Doxepin
Oral.
Mild-medium intensity processes: initial: 25 mg/day at bedtime. Maintenance: 50-150 mg/day in 2-3 doses.
Serious processes: up to max. 300 mg/day.
Contraindications
- Hypersensitivity to this active ingredient.
- Elderly people with acute angle glaucoma or tendency to urinary retention.
Warnings and precautions
- Heart failure and prostatic hypertrophy.
- Cardiac arrhythmias.
- Elders.
- Patients with intercurrent disease or anticholinergic medication.
Interactions with Doxepin
- Enhancement with: central depressants, alcohol.
- Power action of: direct-acting sympathomimetic amines.
- Blocks action of: sympathomimetic amines of indirect action.
- Interrupt MAOIs 2 weeks before starting treatment.
Pregnancy

Avoid. Not recommended.

May cause drowsiness.
Adverse reactions and side effects of Doxepin
Dry mouth.
Blurred vision.
Constipation.
